**4.4.3.1 General information [#w28a151c]

Calcification tissues such as bone, teeth and scale can be detected by von kossa staining procedure.

**4.4.3.2 Reagents and equipments [#a9944fb5]

-5% AgNO3
-Sodium Carbonate-formaldehyde (5% Na2CO3, 10% formaldehyde)
-Farmer’s reducer (10% sodium thiosulfate, 0.5% potassium ferrocyanide)
-Glass slide

**4.4.3.3 Method [#s2207ee0]

+Place sections on a drop of D.W. on to a glass slide.
+Stretch the section and let the section dry at r.t.
+Add a drop of 5% AgNO3 on to the sections and incubate at R.T. for 5~10min in the humid box.
+Rinse 3 times with D.W. for 1 min at R.T.
+Add a drop of Sodium Carbonate-formaldehyde on to the sections and incubate at R.T. for 2min.
+Rinse 3 times with D.W. for 1 min at R.T.
+ Add a drop of Farmer’s reducer on to the sections and incubate at R.T. for 30sec.
+Rinse 3 times with D.W. for 1 min at R.T.
