11.1.2 Acridine orange staining

Acridine orange (AO) is a vital dye that stains apoptotic cells, but not necrotic cells.

11.1.2.1 Reagents

  • 5 µg/ml acridine orange (AO) solution
  • medaka ringer solution

11.1.2.3. Method

  1. Larva or adult fish are incubated in AO solution for 30 to 60 min in the dark.
  2. Wash several times with medaka ringer solution.
  3. Signals are detected using the green (FITC) filter set with a fluorescence microscope.
